Type I Hypersensitivity: IgE-Mediated Responses:

When we model immediate allergic reactions, we focus on robust functional cellular assays that measure basophil and mast cell activation.

Basophil Activation Test (BAT):

A high-sensitivity flow cytometry assay quantifying the upregulation of CD63 and CD203c to evaluate basophil activation following drug or allergen stimulation.

Mast Cell Degranulation:

Functional assays utilizing mature mast cells sensitized with IgE to measure antigen-driven activation and morphological degranulation.

Mediator Release Profiling:

Precise quantification of pre-formed mediators (histamine) and newly synthesized cytokines (IL-4, IL-3) or lipid metabolites (LTC4) via ELISA, fluorometry, or spectrophotometry.

Type IV Hypersensitivity: Sensitization Models

To assess skin sensitization and drug-induced delayed hypersensitivity, we use predictive in vitro models that evaluate dendritic cell maturation and T-cell activation.

T-Cell Priming Assay (TCPA):

Single-cell evaluation of chemical-specific naive human T-cells, measuring proliferation and IFN-γ production.

Myeloid U937 Skin Sensitization Test (MUSST):

An in vitro dendritic cell activation model utilizing the U937 cell line to measure CD86 induction via flow cytometry.

Human Cell Line Activation Test (h-CLAT):

Flow cytometry-based quantification of CD86 and CD54 surface markers on THP-1 cells to effectively discriminate sensitizing drug candidates.
