ELISpot: Vaccine Immunogenicity
For vaccines, the most critical question is not just whether an immune response is generated — but how strong, how specific, and how frequent antigen-responsive cells are at a single-cell level. The ELISpot is a gold-standard method to quantify these responses with high sensitivity.
At ImmunitasBio, we use ELISpot to measure antigen-specific T and B cell responses, enabling precise evaluation of vaccine-induced immunity across preclinical and translational studies.
Single-Cell Resolution of Immune Response
ELISpot detects cytokine or antibody secretion from individual activated cells, where each spot represents a single responsive cell. This allows direct quantification of frequency of antigen-specific immune cells, rather than bulk signal averaging.
We perform assays using PBMCs or isolated immune subsets stimulated with:
- Vaccine antigens or peptide pools
- Recombinant proteins
- Control mitogens and reference standards
Key Readouts for Vaccine Evaluation
T-cell Immunity (Cell-Mediated Response) :
- IFN-γ ELISpot (Th1 response, critical for viral and cancer vaccines)
- IL-2, TNF-α for functional profiling
- Multi-cytokine panels for polyfunctional T-cell assessment
B-cell / Humoral Immunity :
- Detection of antigen-specific antibody-secreting cells
- Memory B-cell response following stimulation
